Modification of Sorbitol by Transglycosylation using Bacillus stearothermophilus Maltogenic Amylase

Abstract

|
Sorbitol was modified by transglycosylation reaction of Bacillus stearothermophilus maltogenic amylase (BSMA). Maltotriose and sorbitol were reacted as a donor and an acceptor, respectively, to produce the sorbitol transglycosylation products. The presence of the sorbitol transfer products was confirmed by HPIC and TLC analyses. The major transglycosylation product was purified by the Bio-Gel P-2 chromatography, and the structure of the major transglycosylation product was determined by MALDI-TOF/ MS, and ^1H- and ^13C-NMR. The major transglycosylation product was determined to be 6^G-¥á-D-maltosyl-sorbitol, in which the maltose unit was attached to the sorbitol by an ¥á-(1 ,6)-glycosidic linkage. The water sorption characteristics of the transfer product show that it has slightly lower water activity than. The water activity of the transfer produt was lower than that of sorbitol at a_w 01-0.7.
